TraDIS-Xpress

TraDIS-Xpress

TraDIS-Xpress (transposon-directed insertion-site sequencing) is a high-resolution whole-genome assay that identifies which genes are likely to affect bacterial fitness in any given condition. We use extremely dense, highly saturated mutant libraries to identify fitness differences at base pair resolution.

TraDIS-Xpress improves upon conventional transposon mutagenesis approaches through our use of outward-facing promoters. This allows the investigation of how gene expression, as well as gene deletion, affects bacterial fitness under a given growth condition. Additionally, it allows the investigation of essential genes that cannot be inactivated and therefore cannot be assayed by other approaches.

At the Quadram Institute, the TraDIS-Xpress team has developed a suite of highly saturated transposon mutant libraries in a range of organisms of interest, which is commercially available through QIB Extra.

We have previously used TraDIS-Xpress to identify:
  • How bacteria respond to an antimicrobial compound
  • How bacterial weaknesses can be exploited to prevent bacterial contamination
  • Mechanisms to improve bacterial fitness in culturing conditions
  • How to increase the production of a protein or metabolite by a host bacteria

What can you expect from TraDIS-Xpress?

  • The QIB Extra TraDIS-Xpress team will meet the client for a discussion about the many benefits of TraDIS-Xpress for your project.
  • The team will design experiments together to suit the clients project requirements and expectations.
  • The experiments carried out by the team will be conducted at the Quadram Institute.
  • The raw and analysed data will be delivered to the client, and discussed in a follow-up meeting.
For a further explanation of how TraDIS-Xpress works, please see the video below from the Quadram Institute, narrated by Prof. Ian Charles OBE of the Quadram Institute:
